- Fluvoxamine contraindicated
in patients with a history of hypersensitivity to the active ingredient.
In patients receiving fluvoxamine therapy, coadministration of
thioridazine, terfenadine, astemizole, cisapride, pimozide, alosetron,
or tizanidine is contraindicated. Fluvoxamine is a potent inhibitor
of several cytochrome P (CYP) isoenzymes, including CYP 1A2, whereas
tizanidine is a CYP 1A2 substrate. In a drug interaction study
of 10 healthy subjects receiving 100 mg of fluvoxamine daily for
four days, administration of a single 4-mg dose of tizanidine resulted
in significantly increased tizanidine maximal (approximately 12-fold;
range, 5- to 32-fold) and mean (33-fold; range, 14- to 103-fold)
plasma concentrations, and an almost threefold prolongation of
its elimination half-life.

- Hyzaar, (losartan potassium plus hydrochlorothiazide), allowing its use to reduce the risk of stroke in patients with hypertension (HTN) and left ventricular hypertrophy (LVH). There is evidence, however, that this benefit does not apply to black patients.
- Panzem, (2-methoxyestradiol capsules - 2ME2) approved as orphan drug indication for use in the treatment
of ovarian cancer.
- Decapinol oral rinse for the treatment of gingivitis in patients aged 12 years and older in whom routine oral
hygiene is not adequate to prevent the inflammatory disease. It is intended for use in conjunction with normal brushing and flossing.
- Boostrix (combination tetanus toxoid, reduced diphtheria toxoid, and acellular pertussis booster vaccine) indicated for administration as a single dose to individuals aged 10 to 18 years old
- Byetta (exenatide) adjunctive therapy to improve glycemic control in patients with type 2 diabetes mellitus
who have not achieved adequate control using metformin and/or a sulfonylurea. Exenatide is an incretin mimetic that enhances glucose-dependent insulin
secretion by pancreatic beta cells, suppresses inappropriately elevated glucagon secretion, and slows gastric emptying.
- Kaletra (lopinavir plus ritonavir) allowing in therapy-naive adults a once-daily dose in combination with
other antiretroviral agents for the treatment of HIV infection.

- Betaseron (interferon
beta-1b) post-marketing safety surveillance reports of serious hepatic injury including autoimmune hepatitis and severe liver damage leading
to hepatic failure and transplant. Liver function testing is recommended at regular intervals.
- Xigris (drotrecogin
alfa - activated) Enrollment in a randomized, double-blind, placebo-controlled trial of Xigris in pediatric patients with severe sepsis was stopped.
Xigris is not indicated for use in pediatric severe sepsis. A planned interim analysis showed that Xigris was highly unlikely to show an
improvement over placebo.

- Lotronex (alosetron) contraindications
and warnings. Concomitant use of alosetron with fluvoxamine is contraindicated.
Fluvoxamine is a known strong inhibitor of CYP1A2 that has been shown
in clinical studies to increase mean alosetron plasma concentrations
by approximately sixfold and prolong its half-life by approximately
threefold.
- Cancidas (caspofungin) should be administered
concomitantly with cyclosporine only in those patients for whom the
potential benefit outweighs the potential risk of elevated liver enzyme
levels.
- Tenoretic (atenolol plus chlorthalidone)
to warn that the atenolol component is associated with a risk of clinically
significant bradycardia in neonates born to mothers administered the
combination drug at parturition or while breast-feeding.
